rClone30-interleukin-12-interleukin-2 is a genetically engineered oncolytic Newcastle disease virus (NDV) strain (Clone 30) designed to express the immunostimulatory cytokines interleukin-12 (IL-12) and interleukin-2 (IL-2). This therapeutic approach combines the direct oncolytic properties of the NDV with the localized production of cytokines to enhance the systemic antitumor immune response. IL-12 promotes Th1 differentiation and activates NK and T cells, while IL-2 supports T-cell proliferation and survival. Preclinical studies in murine hepatoma models demonstrated that the dual-expression vector significantly inhibits tumor growth, increases the secretion of interferon-gamma (IFN-γ) and IP-10, and improves survival rates compared to viruses expressing either cytokine alone.
