Overview

RCUR-313 is an orally bioavailable, potent, and selective small molecule inhibitor of **Lysine-specific demethylase 5B (KDM5B)**, also known as JARID1B. KDM5B is an epigenetic enzyme that removes methyl groups from lysine 4 of histone H3 (H3K4), a modification typically associated with active gene transcription. Overexpression of KDM5B is observed in various malignancies, including breast cancer, melanoma, and lung cancer, where it promotes tumor progression, maintains cancer stem cell populations, and contributes to therapy resistance by silencing tumor suppressor genes. By inhibiting KDM5B, RCUR-313 aims to restore H3K4 methylation levels, thereby reactivating silenced genes, inducing cell cycle arrest, and inhibiting tumor cell proliferation. Originally developed by Recurve Bio, the asset was acquired by Jazz Pharmaceuticals in 2022 and is currently being evaluated in Phase 1 clinical trials for the treatment of advanced solid tumors.
