RD118 is an autologous chimeric antigen receptor T-cell (CAR-T) immunotherapy developed by IASO Bio (Nanjing IASO Biotechnology). It specifically targets G protein-coupled receptor class C group 5 member D (GPRC5D), a surface antigen highly expressed on multiple myeloma cells but limited in normal tissues to plasma and hair follicle cells. The therapy uses a fully human single-domain antibody for high affinity and specificity, with intracellular signaling domains comprising 4-1BB (CD137) and CD3ζ. Its mechanism involves the recognition and elimination of malignant plasma cells expressing GPRC5D, offering a novel approach for patients with relapsed/refractory multiple myeloma, including those who have failed or relapsed after BCMA-targeted therapies. Preclinical studies demonstrated strong cytotoxicity against tumor cells, good expansion/persistence in vivo, and promising safety/efficacy profiles in early clinical trials[2][4][1].
