RDEA806 is an orally active small molecule non-nucleoside reverse transcriptase inhibitor (NNRTI) developed for the treatment of HIV-1 infection. It exhibits potent *in vitro* inhibitory activity against both wild-type and NNRTI-resistant strains of HIV-1, with a high genetic barrier to resistance and a broad spectrum of antiviral activity. Preclinical and clinical studies have shown that RDEA806 has favorable pharmacokinetics, including a terminal half-life up to 13 hours, allowing for once-daily or twice-daily dosing regimens. The drug demonstrates limited potential for drug-drug interactions due to minimal effects on major cytochrome P450 enzymes. In addition to its antiviral properties, its major metabolite (RDEA594) does not possess antiviral activity but lowers serum uric acid levels; as such, development was also considered for gout. The primary developer is Ardea Biosciences.
