Drug intelligence / Profile preview
rDEN3Delta30 (also written as rDEN3Δ30) is a recombinant live attenuated dengue virus serotype 3 (DENV-3) strain originally developed by the National Institute of Allergy and Infectious Diseases (NIAID) of the National Institutes of Health (NIH) as a vaccine candidate. It was constructed by introducing a contiguous 30-nucleotide deletion (Δ30) in the 3' untranslated region (3'-UTR) of the wild-type DENV-3 Sleman/78 strain. Although intended as a vaccine, clinical and preclinical evaluations showed that rDEN3Delta30 was under-attenuated and too reactogenic for use as a vaccine. Consequently, it has been repurposed as a standardized challenge virus in Controlled Human Infection Model (CHIM) studies to evaluate the efficacy of other investigational dengue vaccines (such as VLP Therapeutics' VLPDEN-02) and antiviral therapeutics (such as Janssen's mosnodenvir).
