Overview

rDEN4delta30 (rDEN4Δ30) is a live attenuated viral vaccine candidate developed by the National Institute of Allergy and Infectious Diseases (NIAID) for the prevention of dengue fever caused by dengue virus serotype 4 (DENV-4). The vaccine is engineered using recombinant DNA technology to incorporate a specific 30-nucleotide deletion in the 3' untranslated region (UTR) of the DENV-4 genome, a modification that attenuates the virus's virulence while preserving its immunogenicity. Clinical trials in healthy flavivirus-naïve adults have demonstrated that the vaccine is safe and capable of inducing robust neutralizing antibody responses. In addition to its potential as a monovalent vaccine, rDEN4delta30 serves as a foundational component of NIAID's tetravalent dengue vaccine (TV003/LATV) and as a genetic backbone for chimeric vaccine candidates targeting other dengue serotypes, such as rDEN2/4Δ30.
