Overview

RDHSV-CA8 is a replication-defective herpes simplex virus (rdHSV) based gene therapy developed by Adolore BioTherapeutics for the treatment of chronic pain, specifically targeting knee osteoarthritis. The therapy utilizes a non-integrating viral vector to deliver the Carbonic Anhydrase 8 (CA8) gene to sensory neurons. CA8 is a protein that modulates intracellular calcium signaling by inhibiting the Inositol 1,4,5-trisphosphate receptor type 1 (IP3R1), thereby reducing neuronal hyperexcitability and providing long-lasting analgesia. This approach aims to provide a non-opioid, localized treatment for chronic pain without the risks associated with systemic treatments. The program has received funding from the NIH HEAL Initiative to support development through first-in-human trials, which are expected to commence in 2027.
