RDP-09024

RDP-09024 is an orally bioavailable, non-covalent small-molecule degrader of the c-Myc protein, developed by RDP Pharma AG using its Prompt Degrader™ platform. Unlike conventional PROTACs or molecular glues, RDP-09024 functions by stabilizing a non-native, transiently ordered conformation of the c-Myc homodimer, which triggers protein degradation through both the ubiquitin-proteasome system and aggrephagy-like pathways. Identified as a scaffold-divergent analogue of RDP-02004, RDP-09024 exhibits potent c-Myc degradation and selective cytotoxicity across various MYC-dependent cancer cell lines, including multiple myeloma, lung cancer, and triple-negative breast cancer. It demonstrates an improved off-target profile compared to earlier compounds in the series and has shown ex vivo activity in patient-derived CD138+ multiple myeloma cells and in vivo efficacy in xenograft models.
