RE01 is a small molecule that functions as a positive modulator of Kv3.1 and Kv3.2 voltage-gated potassium channels. This modulation leads to faster activation kinetics and an increased firing frequency in fast-spiking GABAergic interneurons. By enhancing the activity of these channels, RE01 has shown potential in counteracting pathological neuronal network disruptions, such as those observed in Alzheimer's disease where it helps rescue amyloid-beta-induced desynchronization of gamma oscillations. It also demonstrates a capacity to rescue electrophysiological deficits associated with mutated Kv3.1 channels in Progressive Myoclonus Epilepsies, suggesting its therapeutic utility in conditions characterized by impaired neuronal excitability and cognitive dysfunction, including epilepsy, hearing disorders, and schizophrenia.
