SA001 (rebamipide mofetil) is an oral prodrug of rebamipide developed by Samjin Pharmaceutical. It features an ester pro-moiety designed to enhance systemic exposure and bioavailability compared to conventional rebamipide formulations. Rebamipide itself is known for stimulating mucin secretion from goblet cells and modulating inflammation, making it useful in conditions like dry eye disease and Sjögren's syndrome. SA001 has demonstrated greater efficacy than topical rebamipide in preclinical studies for increasing mucin secretion and suppressing pro-inflammatory cytokines such as IL-1, IL-6, and IFN-γ, as well as T cell activation in ocular tissues. Clinical trials have evaluated its safety, tolerability, pharmacokinetics, and efficacy primarily for dry eye symptoms associated with primary Sjögren's syndrome[1][3][4][5].
