REC-1245 is a novel, orally administered small molecule molecular glue developed by Recursion Pharmaceuticals. It selectively induces the degradation of the RNA-binding protein RBM39 via recruitment to the E3 ligase adaptor DCAF15, leading to disruption of RNA splicing and downregulation of cell cycle checkpoints and DNA damage response (DDR) networks, including CDK12. This mechanism mimics the phenotype associated with CDK12 loss-of-function and represents a potential first-in-class approach for modulating DDR pathways in cancer. REC-1245 is being investigated as both a monotherapy and in combination with other agents (such as DDR inhibitors, checkpoint inhibitors, or chemotherapy) for biomarker-enriched solid tumors and lymphoma. The drug was discovered using Recursion’s AI-enabled drug discovery platform and is currently in Phase 1/2 clinical trials[1][3][4][6].
