REC607 (also known as REC-64151) is an orally bioavailable, potent, and selective small molecule inhibitor of the Mucosa-Associated Lymphoid Tissue Lymphoma Translocation Protein 1 (MALT1) protease. Developed by Recursion Pharmaceuticals using their proprietary AI-enabled phenomics platform, the compound is designed to treat various B-cell malignancies and solid tumors by disrupting the NF-κB signaling pathway. MALT1 is a key component of the CBM (CARMA1-BCL10-MALT1) complex, which is essential for transmitting signals from cell surface receptors to the NF-κB transcription factor, a major driver of cell survival, proliferation, and immune response. By inhibiting MALT1's proteolytic activity, REC607 prevents the cleavage of negative regulators of NF-κB, thereby suppressing oncogenic signaling. In addition to its primary focus in oncology, REC607 is being investigated for potential applications in neuroinflammatory and autoimmune disorders where MALT1 activity contributes to pathological immune activation.
