Overview
recMAGE-A3 + AS15 ASCI is an investigational antigen-specific cancer immunotherapeutic (ASCI) consisting of a recombinant MAGE-A3 protein and the AS15 adjuvant system. MAGE-A3 is a tumor-specific antigen expressed in various malignancies, including muscle-invasive bladder cancer, non-small cell lung cancer, and melanoma, but is absent in normal tissues except for the testis and placenta. The AS15 adjuvant system is a potent combination of QS-21, monophosphoryl lipid A (MPL), and CpG 7909 in a liposomal formulation, designed to stimulate both cellular (T-cell) and humoral (antibody) immune responses against MAGE-A3-expressing tumor cells. Developed by GSK, this immunotherapy was evaluated in several Phase II and III trials. However, development was largely discontinued following the failure of Phase III trials in melanoma and lung cancer to meet their primary endpoints.
