recMAGE-A3 + AS15 ASCI

recMAGE-A3 + AS15 ASCI (Antigen-Specific Cancer Immunotherapeutic) is an investigational cancer vaccine developed by GSK. It consists of a recombinant MAGE-A3 protein, which serves as a tumor-specific antigen, combined with the AS15 Adjuvant System. AS15 is a potent immunostimulant containing monophosphoryl lipid A (MPL, a TLR4 agonist), CpG 7909 (a TLR9 agonist), and QS-21 (a saponin-based adjuvant). This combination is designed to induce robust CD4+ and CD8+ T-cell responses as well as antibody responses against MAGE-A3-expressing tumor cells. MAGE-A3 is frequently expressed in melanoma and other cancers but is absent in normal tissues (except testis), making it a highly specific target. Clinical evaluation, including a pilot study led by Craig L. Slingluff, Jr. at the University of Virginia, focused on patients with resected stage IIB-IV MAGE-A3+ melanoma to assess safety and immunogenicity across different administration routes.
