recMAGE-A3 recombinant protein

recMAGE-A3 recombinant protein (also known as ProtD-MAGE-A3/His) is an investigational antigen-specific cancer immunotherapeutic (ASCI) developed by GSK. It consists of a recombinant fusion protein containing the full-length MAGE-A3 tumor-specific antigen, a portion of Protein D from *Haemophilus influenzae*, and a polyhistidine tail. The vaccine is typically formulated with GSK's proprietary AS15 adjuvant system, which includes MPL, QS-21, and CpG 7909 to stimulate a robust T-cell and antibody response against MAGE-A3-expressing tumor cells. MAGE-A3 is a cancer-testis antigen frequently expressed in various malignancies but absent in normal adult tissues. Despite promising early-phase results, the program faced significant setbacks in Phase III clinical trials, including the MAGRIT trial in non-small cell lung cancer and the DERMA trial in melanoma, leading to the discontinuation of development for those indications.
