Drug intelligence / Profile preview
Recombinant adenovirus-herpes simplex virus thymidine kinase (ADV-TK) is an adenoviral vector-based gene therapy developed by Wuhan Tiandakang Biotechnology and Shenzhen Tiandakang Gene Engineering. It is designed to deliver and express the herpes simplex virus thymidine kinase (HSV-TK) gene directly within tumor cells. Following vector administration, the patient is treated with a prodrug such as ganciclovir or valacyclovir. The expressed HSV-TK enzyme phosphorylates the prodrug into a cytotoxic triphosphate metabolite that inhibits DNA polymerase, terminating DNA synthesis and selectively inducing apoptosis in transduced cancer cells. This cytotoxic effect also extends to neighboring untransduced tumor cells via a local bystander effect. The therapy has been evaluated in clinical trials for hepatocellular carcinoma, head and neck cancer, glioma, and nasopharyngeal carcinoma.
