Overview

Recombinant cathepsin G (rCTSG) is a laboratory-produced serine protease that mimics the activity of endogenous cathepsin G, an enzyme typically found in the azurophil granules of neutrophils. In the context of oncology research, specifically breast cancer bone metastasis, rCTSG is investigated for its role as a regulator of tumor plasticity and the bone-tumor interface. Experimental evidence indicates that treatment with rCTSG can reduce breast cancer cell proliferation and induce cell cycle arrest, while simultaneously promoting homotypic aggregation, collective migration, and a luminal-to-basal phenotypic shift. Although it has shown potential in suppressing primary tumor growth in vivo, it paradoxically appears to enhance osteoclast activation and subsequent bone destruction, highlighting its complex role in metastatic progression.
