Overview

rCD95L is a recombinant form of the CD95 ligand (FasL), a type II transmembrane protein belonging to the tumor necrosis factor (TNF) superfamily. It acts as a potent inducer of apoptosis by binding to its cognate receptor, CD95 (Fas/APO-1). Upon binding, rCD95L promotes the formation of the death-inducing signaling complex (DISC), which recruits FADD and pro-caspase-8, leading to the activation of the extrinsic apoptotic cascade. In oncology research, particularly in multiple myeloma, rCD95L is utilized as a tool to investigate the sensitivity of cancer cells to death receptor-mediated apoptosis and to evaluate how factors within the bone marrow microenvironment (BMM) confer resistance to therapies like CAR-T cells that utilize the CD95/CD95L axis for cytotoxicity.
