Overview

Recombinant CHRDL2 (rCHRDL2) is a recombinant form of the chordin-like 2 protein, which functions as a bone morphogenetic protein (BMP) antagonist. It is naturally secreted by the intestinal stem cell (ISC) niche, specifically by subepithelial myofibroblasts and smooth muscle cells. In the context of inflammatory bowel disease (IBD), CHRDL2 is upregulated and acts to promote intestinal epithelial repair. Its mechanism of action involves binding to and activating the epidermal growth factor receptor (EGFR), which subsequently triggers the MAPK signaling pathway and the MUC5B/β-catenin axis. This activation drives the proliferation and regeneration of intestinal stem cells, helping to restore the mucosal barrier. Preclinical studies have demonstrated that administration of rCHRDL2 can alleviate symptoms in DSS-induced colitis mouse models.
