Overview

Recombinant CTHRC1 (rCTHRC1) is a bioengineered version of the Collagen Triple Helix Repeat-Containing 1 protein, a secreted factor involved in tissue repair and highly expressed in various malignant tumors, including pancreatic cancer. In the tumor microenvironment, CTHRC1 acts as a potent endothelial activator that promotes angiogenesis. It functions by inducing the upregulation of angiopoietin-2 (Ang-2) in endothelial cells through ERK-dependent activation of the AP-1 transcription factor. This molecular cascade facilitates the recruitment of Tie2-expressing monocytes (TEMs) to tumor tissues, which supports neovascularization and tumor progression. While rCTHRC1 is primarily used as a research tool to model these pro-tumorigenic pathways, it serves as a critical target for the development of therapeutic strategies, such as neutralizing antibodies, aimed at inhibiting tumor growth and vascularization in CTHRC1-overexpressing cancers.
