Recombinant CXCL9 (rCXCL9) is a biotechnologically produced version of the C-X-C motif chemokine ligand 9, a member of the CXC chemokine family. It functions as a selective agonist for the CXCR3 receptor, which is primarily expressed on activated Th1 cells, CD8+ effector T cells, and natural killer (NK) cells. By activating CXCR3 signaling, rCXCL9 promotes the chemotactic recruitment of these effector immune cells into specific tissues, such as the tumor microenvironment or sites of inflammation. In preclinical research, rCXCL9 has been investigated as an adjuvant to enhance the efficacy of BRAF/MEK inhibitors and anti-PD-1 therapies by increasing T-cell infiltration into "cold" tumors. It has also been studied for its roles in skin fibrosis, hematopoietic stem cell engraftment, and the modulation of myeloid-derived suppressor cells.
