Overview

Recombinant Epstein-Barr Virus Vaccine refers to a class of investigational vaccines designed to prevent or treat infection by the Epstein-Barr virus (EBV), a member of the herpesvirus family that is associated with infectious mononucleosis and several cancers (including nasopharyngeal carcinoma, natural killer T-cell lymphoma, gastric cancer) as well as autoimmune diseases such as multiple sclerosis. These vaccines use recombinant technology to express key EBV antigens—such as gp350, gH/gL/gp42—either via mRNA platforms or nanoparticle display systems. The leading candidates include MDX2201 (developed by ModeX Therapeutics/OPKO Health in collaboration with Merck), which uses a ferritin nanoparticle platform displaying four viral proteins involved in host cell entry to stimulate broad protective immunity against both B cell and epithelial cell infection[2][3][6]. Another candidate is WGc-043 (developed by WestGene Biopharma), an mRNA-based therapeutic cancer vaccine targeting EBV-related malignancies[1][4][5]. As of June 2025, no recombinant EBV vaccines are approved for use; all are in early clinical development.
