Recombinant extracellular nicotinamide phosphoribosyltransferase (eNAMPT) is a laboratory-produced version of the secreted form of the NAMPT enzyme, also known as visfatin or pre-B-cell colony-enhancing factor (PBEF). While intracellular NAMPT (iNAMPT) is the rate-limiting enzyme in the NAD+ salvage pathway, the extracellular form (eNAMPT) functions as a potent pro-inflammatory cytokine and adipokine. Recombinant eNAMPT is primarily used as a research tool to investigate its role in inflammatory pathologies, such as Crohn's disease and other forms of inflammatory bowel disease (IBD). It functions as an endogenous ligand and agonist for Toll-like receptor 4 (TLR4), triggering signaling pathways that promote the expansion of myeloid progenitors in the bone marrow and the infiltration of immature granulocytes into inflamed tissues. In experimental models, administration of recombinant eNAMPT exacerbates intestinal inflammation and fibrotic remodeling, making it a key target for neutralization strategies in chronic inflammatory conditions.
