Recombinant Fas Ligand (FasL) is a protein belonging to the tumor necrosis factor (TNF) superfamily that functions as a potent inducer of apoptosis. It acts by binding to and trimerizing the Fas receptor (CD95), which triggers the extrinsic apoptotic pathway via the recruitment of FADD and activation of caspase-8. While primarily used as a research reagent to study programmed cell death and immune evasion (as seen in glioblastoma models), recombinant versions of FasL have been investigated as potential antineoplastic agents. However, clinical development has been challenged by systemic toxicity, particularly hepatotoxicity, leading to the engineering of specialized variants like hexameric fusion proteins to improve safety and efficacy.
