Recombinant flt3 ligand (Flt3L) is a hematopoietic growth factor that plays a critical role in the proliferation and differentiation of early hematopoietic progenitor cells and dendritic cells. In a Phase I clinical trial (NCT00003843) conducted by Northwestern University, this recombinant protein was used as a key component of an ex vivo cytokine cocktail—alongside interleukin-3 (IL-3), G-CSF, GM-CSF, and erythropoietin—to expand peripheral blood mononuclear cells (PBMNCs) harvested from patients with relapsed or refractory non-Hodgkin's lymphoma. The therapeutic objective was to re-infuse these expanded cells following high-dose chemotherapy to accelerate bone marrow recovery and eliminate the period of severe neutropenia. Flt3L functions by binding to the FLT3 receptor (CD135), a tyrosine kinase receptor expressed on the surface of multipotent hematopoietic progenitors.
