Overview

Recombinant gp100 protein is a cancer vaccine candidate developed primarily by the National Cancer Institute (NCI) for the treatment of melanoma. It consists of a laboratory-produced version of the gp100 (also known as PMEL or PMEL17) protein, a lineage-specific differentiation antigen that is highly expressed in melanocytes and melanoma cells. As an immunotherapeutic agent, the protein serves as an antigen that is taken up, processed, and presented by antigen-presenting cells (APCs) to the immune system. This process is intended to activate and expand gp100-specific CD4+ T-helper cells and CD8+ cytotoxic T lymphocytes, thereby generating a targeted immune response against melanoma tumor cells. Clinical investigations have explored various formulations, including emulsification with adjuvants such as Montanide ISA-51, the use of modified variants like the 184V protein to enhance immunogenicity, and the loading of the protein onto dendritic cells for ex vivo stimulation.
