Drug intelligence / Profile preview
Recombinant GPNMB (glycoprotein nonmetastatic melanoma protein B), also known as recombinant osteoactivin, is a recombinant protein formulation of the extracellular domain of GPNMB. It is being developed as a therapeutic agent by GPN Therapeutics, a spinout from Northeast Ohio Medical University (NEOMED), for the treatment of musculoskeletal and degenerative disorders, including post-traumatic osteoarthritis, osteoporosis, and bone regeneration in spinal fusion. GPNMB is an anti-inflammatory glycoprotein that acts as an agonist of the CD44 receptor, promoting mesenchymal stem cell recruitment, osteoblast differentiation, and M2 macrophage polarization while suppressing neuroinflammation and cartilage degradation. In preclinical models, intra-articular administration of recombinant GPNMB has demonstrated chondroprotective and subchondral bone protective effects, making it a promising disease-modifying candidate for osteoarthritis.
