Overview

Recombinant HAPLN1 (rHAPLN1) is a recombinant form of the Hyaluronan and Proteoglycan Link Protein 1, a secreted extracellular matrix (ECM) protein. HAPLN1 is essential for maintaining the structural integrity of the ECM by stabilizing the binding of hyaluronan to chondroitin sulfate proteoglycans. Research, particularly in the context of aging and oncology, has demonstrated that age-dependent loss of HAPLN1 in the dermis leads to increased matrix stiffness. This physical change in the tumor microenvironment triggers the upregulation of endothelial ICAM1 and the downregulation of VE-cadherin, which erodes vascular integrity and promotes melanoma metastasis. Preclinical studies have shown that intradermal treatment with rHAPLN1 can restore vascular barrier function and reduce the metastatic potential of melanoma cells by normalizing the aged ECM.
