Recombinant hemagglutinin (rHA) influenza H5N1 vaccines are a class of subunit vaccines designed to provide active immunization against the H5N1 subtype of the Influenza A virus, commonly known as avian influenza. These vaccines are produced using recombinant DNA technology, typically involving the expression of the viral hemagglutinin (HA) protein in cell culture systems such as insect cells (via baculovirus vectors) or mammalian cells, rather than traditional embryonated chicken eggs. The HA protein is the primary target for neutralizing antibodies, as it facilitates viral attachment and entry into host cells. By delivering purified rHA, these vaccines elicit an immune response specific to the H5N1 surface antigen. This technology allows for a faster manufacturing response during a pandemic compared to egg-based methods and avoids issues related to egg allergies or viral adaptation to eggs. The first such vaccine, developed by Protein Sciences (now part of Sanofi), received FDA approval for use in adults at increased risk of exposure to the H5N1 virus.
