Drug intelligence / Profile preview
Recombinant heparanase 2 (rHpa2) is a recombinant form of the human heparanase 2 (Hpa2) protein, a non-enzymatic homolog of heparanase-1 (Hpa1). Unlike Hpa1, Hpa2 lacks endoglycosidase activity but retains a high affinity for heparan sulfate (HS). It acts as an endogenous inhibitor of Hpa1 by competing for HS substrate binding and physically interacting with Hpa1 to prevent HS cleavage. Additionally, rHpa2 competes with heparin-binding growth factors, such as fibroblast growth factor 2 (FGF2) and vascular endothelial growth factor A (VEGFA), for binding sites on the endothelial cell surface, thereby modulating downstream signaling pathways. Developed by researchers at Hannover Medical School and the MDI Biological Laboratory, rHpa2 is being investigated for its potential to maintain vascular integrity, prevent endothelial glycocalyx degradation, and treat microvascular diseases, proteinuria, glomerulonephritis, sepsis, and pulmonary fibrosis.
