Overview

Recombinant HMGB1 (rHMGB1) is a laboratory-produced version of the High-Mobility Group Box 1 protein, a highly conserved nuclear protein that acts as a potent Damage-Associated Molecular Pattern (DAMP) when released into the extracellular environment. It functions as a proinflammatory mediator by binding to several cell-surface receptors, most notably the Receptor for Advanced Glycation End-products (RAGE), Toll-like receptor 2 (TLR2), and Toll-like receptor 4 (TLR4). This binding triggers intracellular signaling pathways, such as NF-κB, leading to the production of inflammatory cytokines including IL-1, IL-6, and IL-8. In dermatological research, rHMGB1 is utilized to study the pathogenesis of inflammatory conditions like psoriasis, where it has been shown to stimulate keratinocytes and fibroblasts to adopt a proinflammatory phenotype. While primarily used as a research reagent to study inflammatory pathways, it is also investigated for its role in tissue repair and various autoimmune diseases.
