Overview

Recombinant human acidic fibroblast growth factor (rh-aFGF) is a disulfide-linked monomeric protein consisting of approximately 141 amino acids with a molecular weight around 16 kDa. It is produced by expressing an optimized DNA sequence encoding the mature chain of human acidic FGF in Escherichia coli or insect cells. This protein belongs to the fibroblast growth factor family and binds to FGF receptors (a family of receptor tyrosine kinases), activating signaling pathways important for cell proliferation and differentiation. Mechanistically rh-aFGF acts as an agonist of fibroblast growth factor receptors promoting DNA synthesis, cell proliferation (including endothelial cells), chemotaxis and angiogenesis. It plays roles in tissue repair processes such as wound healing by stimulating fibroblasts proliferation and new blood vessel formation through ERK1/2 and STAT3 signaling pathways. Clinical studies have shown topical rh-aFGF accelerates healing in deep partial-thickness burns, skin graft donor sites and pressure sores including those caused by facemask use during COVID-19.
