Recombinant human beta-beta-defensin 2 (rHuBβD2) is a recombinant peptide biologic being developed by Planton as a topical cream formulation for the treatment of atopic dermatitis (AD). The active ingredient is a recombinant form of human beta-defensin 2 (HBD-2), a 64-amino acid, cysteine-rich cationic host defense peptide naturally secreted by epithelial cells in response to microbial contact or pro-inflammatory stimuli. HBD-2 possesses both direct antimicrobial properties, which disrupt bacterial cell membranes, and skin barrier-protective functions. Specifically, it inhibits Staphylococcus aureus-secreted proteases (such as V8/SspA) that degrade the skin barrier, addressing the host defense peptide deficiency and S. aureus colonization that worsen AD lesions. Additionally, HBD-2 acts as an agonist of C-C chemokine receptor type 6 (CCR6) and C-C chemokine receptor type 2 (CCR2), promoting chemotaxis of immune cells. Planton has completed a Phase 1 clinical trial in Australia evaluating topical rHuBβD2 cream versus placebo in patients with mild to moderate atopic dermatitis colonized with S. aureus.
