Overview

Recombinant human calcineurin B (rhCNB) is a recombinant protein representing the regulatory B subunit of calcineurin, a calcium-dependent serine/threonine phosphatase. Developed by Haikou Qili Pharmaceutical, rhCNB is being investigated as an antineoplastic agent for the treatment of various solid tumors, most notably gastric cancer and hepatocellular carcinoma. Unlike the complete calcineurin heterodimer, the isolated B subunit has been found to possess intrinsic anti-tumor properties. Its mechanism of action involves binding to Toll-like receptor 4 (TLR4) on the surface of immune and tumor cells, which triggers signaling cascades that lead to the induction of apoptosis in cancer cells and the activation of macrophages and other immune components. rhCNB has demonstrated the ability to inhibit tumor growth and metastasis in preclinical models and is currently undergoing clinical evaluation in China.
