DEL-1 (Developmental Endothelial Locus-1), also known as EDIL3, is a secreted glycoprotein that serves as an endogenous anti-inflammatory factor. It functions primarily by regulating leukocyte recruitment, specifically by antagonizing the adhesion of neutrophils to the vascular endothelium. DEL-1 acts as a gatekeeper of the inflammatory response by binding to the integrin alpha-V beta-3 on endothelial cells, which prevents the interaction between the leukocyte integrin LFA-1 and its ligand ICAM-1. Additionally, DEL-1 can directly interact with LFA-1 on neutrophils to inhibit their activation and recruitment. Recombinant forms of DEL-1 are being investigated as therapeutic agents for inflammatory and autoimmune conditions, including periodontitis and multiple sclerosis, where the protein's ability to promote the resolution of inflammation and prevent tissue destruction is of high clinical interest.
