Overview

rh-ES + CV is a combination therapeutic regimen consisting of recombinant human endostatin (rh-ES), carboplatin, and vincristine. Recombinant human endostatin, marketed under the brand name Endostar, is an endogenous broad-spectrum angiogenesis inhibitor that targets tumor vascularization by interfering with pro-angiogenic signaling pathways. Carboplatin is a platinum-based cytotoxic small molecule that acts as a DNA alkylator, inducing DNA cross-links that inhibit replication and transcription. Vincristine is a vinca alkaloid that functions as a microtubule inhibitor, disrupting the mitotic spindle and causing cell cycle arrest. This combination is primarily being evaluated in clinical trials sponsored by Beijing Sanbo Brain Hospital for the treatment of newly diagnosed pediatric low-grade gliomas (LGG), particularly those that are unresectable or located in critical midline structures such as the visual pathway or brainstem. The rationale for the regimen is that the anti-angiogenic properties of rh-ES may synergize with the cytotoxic effects of the carboplatin and vincristine backbone to improve tumor response and progression-free survival in pediatric patients.
