Recombinant human fibrinogen (rhFIB) is a biosynthetic form of the naturally occurring blood-clotting factor I. It is a complex glycoprotein composed of two sets of three polypeptide chains (alpha, beta, and gamma). Produced using recombinant DNA technology—most notably in the milk of transgenic dairy cows or in mammalian cell lines such as CHO cells—it is designed to serve as a replacement therapy for patients with congenital or acquired fibrinogen deficiencies. The mechanism of action involves the cleavage of rhFIB by thrombin to produce fibrin monomers, which then polymerize to form a stable fibrin clot, restoring hemostasis. It is being investigated for use in treating bleeding disorders and as a component of surgical tissue sealants, offering a potential safety advantage over plasma-derived fibrinogen by eliminating the risk of human viral contamination.
