Recombinant human fibrinogen Aα847 (rhFbgAα847) is an investigational recombinant biologic variant of human fibrinogen being developed by the Leids Universitair Medisch Centrum (LUMC) in Leiden, Netherlands. It is a recombinant form of fibrinogen—a key blood coagulation protein—in which the Aα chain is expressed at an extended length of 847 amino acids, distinguishing it from the wild-type variant (rhFbgAα610). The Aα847 variant is designed to be more resistant to fibrinolysis than wild-type or plasma-derived fibrinogen. It is being investigated for the treatment and management of coagulation disorders and coagulopathy, particularly perioperative bleeding and fibrinogen deficiency in patients undergoing cardiac surgery (such as isolated on-pump coronary artery bypass graft surgery). In the ex vivo FIBRINOGEN study, the blood coagulation effects of rhFbgAα847 are compared with rhFbgAα610 and the plasma-derived fibrinogen concentrate Haemocomplettan using rotational thromboelastometry (ROTEM) on blood samples collected from patients undergoing cardiac surgery.
