Overview

Recombinant human hyaluronidase PH20 (rHuPH20) is a purified recombinant enzyme produced in genetically engineered Chinese Hamster Ovary (CHO) cells containing a DNA plasmid encoding for the soluble fragment of the human hyaluronidase enzyme PH20[1][2][3]. It acts locally to temporarily depolymerize hyaluronan—a major glycosaminoglycan component of the extracellular matrix—by hydrolyzing the N-acetylglucosamine and glucuronic acid bond[4][5]. This enzymatic action increases tissue permeability and facilitates rapid subcutaneous delivery and absorption of large volumes or high doses of co-administered therapeutics, including drugs and fluids[3][4][6]. rHuPH20 is used as an adjuvant to enhance subcutaneous drug delivery and has been shown to be well tolerated with a favorable safety profile. The technology was developed by Halozyme Therapeutics.
