Overview

Recombinant human lymphotoxin-α derivative (rhLTα-Da) is a genetically engineered form of the cytokine lymphotoxin-α (also known as tumor necrosis factor-beta), in which 27 N-terminal amino acid residues are deleted. It is developed as a biological antitumor agent and acts by modulating the activity of TNF receptors (TNFR55 and TNFR75), similar to tumor necrosis factor alpha but with reduced toxicity. The drug has been studied primarily in China for use in combination with chemotherapy regimens such as cisplatin and fluorouracil for metastatic esophageal squamous cell carcinoma. While overall efficacy was not demonstrated across all patients, some benefit was observed in subgroups with low serum TNF receptor II levels. The most common adverse effects include chills and fever[1][2][3][6].
