Overview

Recombinant human phospholipase C zeta (rhPLCζ) is an investigational biologic therapeutic being developed for the treatment of oocyte activation failure (OAF) and male factor infertility. It is a recombinant version of the sperm-specific enzyme PLCζ1, which is responsible for triggering the intracellular calcium (Ca²⁺) oscillations necessary for oocyte activation and subsequent embryo development. In assisted reproduction, rhPLCζ is administered via microinjection during intracytoplasmic sperm injection (ICSI) to couples where the sperm lacks sufficient endogenous PLCζ or where previous fertilization attempts have failed. This approach aims to provide a more physiological stimulus than traditional chemical calcium ionophores. Clinical pilot studies, notably at Weill Cornell Medicine, have indicated that rhPLCζ can successfully rescue fertilization and lead to viable pregnancies in cases of sperm-related activation deficiencies.
