HB-002.1 is a recombinant human VEGF receptor-antibody fusion protein developed by Huabo Biopharmaceutical, a subsidiary of Zhejiang Huahai Pharmaceutical. Designed as a biosimilar to aflibercept, it utilizes a "VEGF Trap" architecture to sequester pro-angiogenic ligands. The molecule is a dimeric glycoprotein consisting of the extracellular immunoglobulin (Ig) domain 2 of human VEGF receptor 1 (VEGFR1) and Ig domain 3 of human VEGF receptor 2 (VEGFR2), fused to the Fc portion of human IgG1. By acting as a high-affinity soluble decoy receptor, HB-002.1 binds to and neutralizes vascular endothelial growth factor A (VEGF-A), VEGF-B, and placental growth factor (PlGF). This mechanism inhibits the activation of endogenous VEGF receptors, thereby suppressing pathological angiogenesis and reducing vascular permeability. It is primarily being developed for ophthalmic conditions, specifically neovascular (wet) age-related macular degeneration (nAMD), where it is administered via intravitreal injection.
