Overview

Recombinant hypoallergenic Bet v 1 derivatives are genetically engineered proteins based on the major birch pollen allergen Bet v 1, produced to reduce their IgE-binding capacity while retaining T-cell immunogenicity. These derivatives—including folding variants (rBet v 1-FV), trimeric forms, and fragments—are designed as safer and effective agents for allergen-specific immunotherapy (AIT) of birch pollen allergy. By modifying the native Bet v 1 sequence or structure, these recombinant derivatives minimize anaphylactic activity while still eliciting immune responses that lead to tolerance or blocking antibody (IgG) production. Multiple clinical trials have shown efficacy in reducing allergic rhinoconjunctivitis symptoms and induction of immune tolerance with an improved safety profile compared to natural extract vaccines. Some derivatives are administered subcutaneously or intranasally, and others are evaluated for sublingual use[1][3][4][6][7].
