The adjuvant enhances humoral and cellular immune responses to the influenza antigen, promoting both Th1 (IgG2a, IFN-γ, IL-2) and Th2 (IgG1, IL-5, IL-6) cytokine profiles, increased CD4/CD8 T-cell proliferation, higher neutralizing antibody titers, and antigen dose-sparing (10-100 fold). It provides superior protection against lethal H5N1 influenza challenge in ferrets compared to unadjuvanted antigen, with sustained immunity up to 12 months and low reactogenicity, outperforming squalene-based adjuvants like MF59 in safety while matching efficacy.
