Overview

**Recombinant interleukin-21 + sunitinib** is an experimental combination therapy comprising recombinant interleukin-21 (rIL-21), a biologic cytokine, and sunitinib, a small molecule multi-targeted receptor tyrosine kinase inhibitor. rIL-21 acts by stimulating the activation and proliferation of CD8+ T cells and natural killer (NK) cells, thereby enhancing antitumor immune responses[1][2][4][7]. Sunitinib inhibits several receptor tyrosine kinases, notably vascular endothelial growth factor receptors (VEGFRs), platelet-derived growth factor receptors (PDGFRs), and other growth factor receptors, resulting in antiangiogenic and antiproliferative effects. Preclinical studies and early clinical trials indicate that combining rIL-21 with tyrosine kinase inhibitors such as sunitinib may boost antitumor efficacy relative to monotherapy, primarily by enhancing both innate and adaptive antitumor immunity and inhibiting tumor angiogenesis[7]. Both agents have been primarily investigated in metastatic cancers, such as renal cell carcinoma (RCC) and melanoma, though direct clinical trials for the specific combination are limited.
