Overview

Recombinant interleukin-33 (IL-33) is a biologic protein that mimics the endogenous IL-33 cytokine, a member of the IL-1 superfamily. It acts as an "alarmin" released from the nucleus of damaged or necrotic cells to alert the immune system. IL-33 functions by binding to its specific receptor, ST2 (also known as IL-1RL1), which is expressed on various immune cells including group 2 innate lymphoid cells (ILC2s), Th2 cells, mast cells, and regulatory T cells. This binding recruits the IL-1 receptor accessory protein (IL-1RAcP) and activates intracellular signaling pathways such as NF-κB and MAP kinases. While primarily utilized as a research reagent, recombinant IL-33 has demonstrated therapeutic potential in preclinical models for promoting tissue repair, enhancing diabetic wound healing, and improving metabolic fitness by activating beige thermogenesis in adipose tissue via the ERα/NAMPT/IL-33 axis.
