Overview

Recombinant KLK10 (rKLK10) is a recombinant form of the Kallikrein-Related Peptidase 10, a secreted serine protease that plays a critical role in vascular health. Research conducted at Emory University has identified KLK10 as a flow-sensitive protein that is upregulated under stable blood flow conditions and significantly downregulated under disturbed flow or in atherosclerotic plaques. In preclinical models of atherosclerosis, administration of rKLK10 has been shown to inhibit endothelial inflammation and reduce the formation of atherosclerotic plaques. Its mechanism of action involves the modulation of Protease-Activated Receptors 1 and 2 (PAR1 and PAR2) and the subsequent inhibition of the NFκB signaling pathway, leading to decreased expression of pro-inflammatory adhesion molecules such as VCAM1 and ICAM1. It is being investigated as a potential therapeutic for atherosclerosis and related cardiovascular inflammatory conditions.
