Overview

Recombinant *Lactococcus lactis* expressing ovalbumin is a genetically engineered live biologic platform used primarily as a research tool in immunology and oncology. The probiotic bacterium *Lactococcus lactis* (often strain MG1363) is modified to express or secrete chicken ovalbumin (OVA), a model antigen. In therapeutic research, it is utilized to induce mucosal immune tolerance for treating allergic or autoimmune diseases or as an adjuvant-antigen delivery vehicle for dendritic cell-based cancer vaccines. The bacterium interacts with the immune system by promoting dendritic cell maturation through the Toll-like receptor 2 (TLR2) pathway and enhancing antigen cross-presentation via the phagosome-to-cytosol pathway. This process is dependent on reactive oxygen species (ROS) production and proteasomal degradation, leading to the activation of antigen-specific CD8+ cytotoxic T lymphocytes and Th1 responses.
