Overview

rLIGHT (recombinant LIGHT) is a recombinant form of the cytokine Tumor Necrosis Factor Superfamily Member 14 (TNFSF14). It acts as a potent agonist for two primary receptors: the Lymphotoxin beta receptor (LTβR) and the Herpesvirus entry mediator (HVEM/TNFRSF14). In the tumor microenvironment, rLIGHT is investigated for its ability to stimulate anti-tumor immune responses and promote the formation of tertiary lymphoid structures. In respiratory research, rLIGHT has been identified as a driver of airway remodeling and hyperresponsiveness (AHR) in asthma models. It directly induces airway smooth muscle cell (SMC) contraction, migration, and hyperplasia by activating canonical and non-canonical NF-κB signaling, which subsequently triggers Rac1 and PAK1 pathways. This leads to actin polymerization and myosin light chain phosphorylation, resulting in actomyosin formation and muscle contraction. While primarily used as a research tool to study asthma pathogenesis, recombinant human LIGHT (rhLIGHT) is also being evaluated in early-phase clinical trials for the treatment of advanced solid tumors.
