Recombinant methioninase is a recombinant enzyme that depletes methionine, an amino acid essential for the survival and proliferation of many cancer cells. The drug targets "methionine addiction," a metabolic hallmark of cancer known as the Hoffman effect, where cancer cells require exogenous methionine due to increased transmethylation reactions. By degrading circulating methionine, recombinant methioninase selectively arrests cancer cell growth and enhances sensitivity to chemotherapy agents. It has demonstrated efficacy in vitro and in vivo against various cancers including lung, colon, kidney, melanoma (both BRAF-V600E-positive and negative), brain, prostate, ovarian, pancreatic cancers, and triple-negative breast cancer.
