The recombinant Newcastle Disease Virus (NDV) vectored vaccine for SARS-CoV-2 is a viral vector vaccine platform designed to prevent COVID-19. It utilizes a modified version of the Newcastle Disease Virus, a poultry pathogen that does not cause disease in humans, to express a stabilized version of the SARS-CoV-2 spike protein (HexaPro). This stabilized spike protein contains six proline substitutions (HXP) that lock it into a prefusion conformation, significantly increasing its stability and immunogenicity compared to first-generation spike antigens. The vaccine can be produced in embryonated chicken eggs, leveraging existing global influenza vaccine manufacturing infrastructure. It is designed for administration via intramuscular injection or intranasal spray, the latter of which aims to induce mucosal immunity in the upper respiratory tract. The technology was primarily developed by the Icahn School of Medicine at Mount Sinai in collaboration with the University of Texas at Austin and Wake Forest University, and has been licensed to several international partners for local production.
